Polyacrylamide Gel Electrohoresis (PAGE) is a powerful tool for purifying oligonucleotides. The separation is mainly based on charge and mass - the longer the molecule the larger the charge and of course the mass.
The PAGE is performed under denaturing conditions and the separation is done with a resolution down to one base.
After PAGE the analysis is visualized by UV shadowing. The desired band is cut out of the gel. After that the oligo is eluted from the gel with water or buffer and subsequently desalted and quantified.
Oligos used in mutagenesis, structural studies etc. are candidates for PAGE purification.
